چکیده

Surface functionalisation with viscosity sensitive dyes termed 'molecular rotors' can potentially open up new opportunities in sensing, for example for non-invasive biological viscosity imaging, in studying the effect of shear stress on lipid membranes and in cells, and in imaging contacts between surfaces upon applied pressure. We have functionalised microscope slides with BODIPY-based molecular rotor capable of viscosity sensing via its fluorescence lifetime. We have optimised functionalisation conditions and prepared the slides with the BODIPY rotor attached directly to the surface of glass slides and through linkers of ∼40 nm and ∼300 nm in length. The slides were characterised for their sensitivity to viscosity, and used to measure viscosity of supported lipid bilayers during photooxidation, and of giant unilamellar vesicles lying on the surface of the slide. We conclude that our functionalised slides show promise for a variety of viscosity sensing applications.

Tuning BODIPY molecular rotors into the red: sensitivity to viscosity vs. temperature

Viscosity variations in the microscopic world are of paramount importance for diffusion and reactions. In the last decade a new class of fluorescent probes for measuring viscosity has emerged termed ‘molecular rotors’, which allows quantitative mapping of viscosity in microscopically heterogeneous environments. Here we attempt to tune the absorption and emission of one such ‘molecular rotor’ ba...

BODIPY-BODIPY dyad: assessing the potential as a viscometer for molecular and ionic liquids.

A symmetrical BODIPY-BODIPY dyad with a diyne linker was prepared in two steps; the lifetime decay of this rotor appeared to correlate with the viscosity of the media, thus making this dyad a suitable small molecule viscometer for molecular solvents. The potential of using the rotor to probe the viscosity of ionic liquids was also investigated.

Molecular rheology of neuronal membranes explored using a molecular rotor: Implications for receptor function.

The role of membrane cholesterol as a crucial regulator in the structure and function of membrane proteins and receptors is well documented. However, there is a lack of consensus on the mechanism for such regulation. We have previously shown that the function of an important neuronal receptor, the serotonin1A receptor, is modulated by cholesterol in hippocampal membranes. With an overall object...

A highly sensitive viscosity probe based on ferrocene-BODIPY dyads.

Two novel ferrocene-BODIPY molecules FcEB and FcB have been synthesized and characterized. An "interlock" packing mode was demonstrated in both FcEB and FcB by analysis of X-ray diffraction data. FcEB exhibits a molecular rotor property with a significant enhancement of quantum yield in viscous solvent. The result shows that the emission of FcEB increased dramatically as the viscosity of soluti...

A molecular rotor for measuring viscosity in plasma membranes of live cells.

Molecular rotors have emerged as versatile probes for microscopic viscosity in live cells, however, the exclusive localisation of rotors in the plasma membrane has remained elusive. We report the synthesis, spectroscopic characterisation and live cell imaging of a new BODIPY-based molecular rotor suitable for mapping viscosity in the cell plasma membrane.
